My wife is self employed and I have a regular job where taxes are taken from my pay and I receive a W-2.

Yes.  If your wife has both self-employment income and expenses, either TurboTax Premier or Self-employed Online Editions would be suitable or if you prefer a desktop version, TurboTax Deluxe or higher version would handle your situation.
