No, you only pay Capital Gains on your Federal Return, or the State the home was located in the year of the sale.
If you lived in your home for two of the last five years, and your gain was less than 250K (if single) or 500K (married), you don't have to pay tax on your gain at all.
In fact, you wouldn't even have to enter the sale of your home in that case, unless you received a 1099-S.
