I started a home daycare this year but also worked in my mother's home daycare as an assistant. Should I include both income amounts under the same line of work?

The work you did as an assistant should be reported on a W2 issued by your mother. That would NOT be included in your self-employment income. 

If you had a different arrangement with your mother and was working as a contract worker (you get a 1099-MISC), then yes, enter that income together with the income you earned at the location in your home.
