I'm not sure by looking at your question. Assuming you are saying you and your spouse live and work in MO and your spouse's employer is based in FL then yes you need to file a MO resident return. You will be taxed by MO on all income from all sources. Since there was nothing withheld for MO taxes by the FL employer you may owe taxes on that income.

Sorry if I was not clear. We are staying in FL and her office is in St Louis MS. The W2 we got did not have any state tax info. Q is do we need to file non resident tax for MS as FL doesn't have state tax which is resident state
Then, no. Your resident state is FL which is where you live and work and no one physically worked in MO so no MO taxes are due. Further, since nothing was withheld by MO, you do not need to file a MO tax return.
