No, since you already set up a payment option with your original return, and your Amended return shows a refund, so you do not have to mail in the payment voucher. TurboTax does not know when a withdrawal for taxes owed hits.

After I e-filed my amended tax return, it gave me a 1040V voucher. It states that I need to mail a check to the IRS. From my original return, I selected April 10th for the payment to be withdrawn. Would I have to mail the check even though I'm getting a refund from the amended return. It's says in Line 18 from the 1040-X form that my overpayment was zero and Line 20 for the amount I owe is blank.

Your original return is processed separate from your amended return. The IRS will still deduct the amount owed from your original return. Your amended return since it shows a refund will be sent to you by check.
