Yes, a 1065 form can only be created in the TurboTax Business software.

Can TT Business upload the data I've already entered in TT SE?

No, the forms in each of the software versions are completely different so there is not any way to transfer the data. The only importing that can happen in TurboTax Business is from Quickbooks.
