I filed my return, I selected my Credit Karma account as my refund method. When my return is approved will I get the refund advance? Or do I sign up for that separately?

The refund advance is something separate than your tax return, and had to be applied for before completing your tax return. 

You can learn more about the refund advance here. Not all tax filers are able to apply for a refund advance.

Thank you! So now there's no way to select that method? With my Credit Karma account as the selected method, will I most likely receive payment within 21 days of approval?

If you were eligible for the Refund Advance, it should have been offered at the time you filed your tax return. If you selected to have your refund deposited into your Credit Karma Money Spend account, you may receive your refund up to 5 days early.
