Married filing separately status changed to single filing status. Do I need to amend?

Were you married at the end of 2021? If you were, then you would use the Married filing status. If you were not still Married at the end of 2021, you can use the Single filing status or if you qualify Head of Household.

If you filed your 2021 return with the wrong filing status, you would amend your return to correct the filing status. You should wait until your 2021 return(s) are fully processed before you amend.
